ACTIDOL-DP - Used for: Treating moderate to moderately severe pain.

ACTIDOL-DP is an analgesic. It works in certain areas of the brain and nervous system to decrease pain.

ACTIDOL-DP is an opioid analgesic, a derivative of cyclohexanol. It is non-selective agonist of mu-, delta- and kappa-receptors in the CNS. ACTIDOL-DP is a racemate (+) and (-) of isomers (50% / 50%) which in various ways are involved in analgesic effects. The isomer (+) is a pure agonist opioid receptors, it has low tropism and has a pronounced selectivity for different subtypes of receptors. The isomer (-) inhibiting neuronal capture of noradrenaline activates the descending noradrenergic influence. Thats why it is broken transmission of pain impulses in the gelatinous substance of spinal cord.

ACTIDOL-DP causes sedation. At therapeutic doses this medication almost not get respiratory depression. ACTIDOL-DP has also an antitussive effect.

ACTIDOL-DP, a centrally-acting analgesic, exists as a racemic mixture of the trans isomer, with important differences in binding, activity, and metabolism associated with the two enantiomers. Although ACTIDOL-DP is a synthetic analog of codeine, it has a significantly lower affinity for opioid receptors than codeine.

Indications:

Adult: PO Moderate to severe pain 50-100 mg 4-6 hrly. Max: 400 mg/day. Extended-release 50-100 mg 1-2 times/day. Max: 300 mg/day. IV/IM Moderate to severe pain 50-100 mg 4-6 hrly. Post-op pain Initial: 100 mg, then 50 mg every 10-20 mins if needed, up to 250 mg for the 1st hr.

Maintenance: 50-100 mg 4-6 hrly. Max: 600 mg/day. Rectal Moderate to severe pain As 100 mg supp: 1 supp up to 4 times/day.

Oral Moderate to severe pain

Adult: As conventional tablet: 50-100 mg every 4-6 hr. Max: 400 mg daily. As extended-release tablet: 50-100 mg once or twice daily. Max: 300 mg daily.

Elderly: Lower initial dose. Max: 300 mg daily (>75 yr). Max

Dosage: 400 mg daily. CrCl (ml/min) Dosage Recommendation 10-<30 As conventional tablet: increase dosage interval to 12 hr; max: 200 mg/day. <10 Contraindicated. Hepatic impairment: As conventional tablet: Increase dosage interval to 12 hr in severe impairment e.g. 50 mg bid in hepatic cirrhosis. As extended-release tablet: Contraindicated in Child Pugh Class C. Parenteral Moderate to severe pain

Adult: IM/IV inj over 2-3 min/IV infusion: 50-100 mg given every 4-6 hr.
