Esoral - Pharmacology: Suppresses gastric acid secretion by blocking proton pump within gastric parietal cells.

Indication: Oral

Short-term treatment of heartburn and other symptoms of gastroesophageal reflux disease (GERD); short-term treatment in healing and symptomatic resolution of erosive esophagitis; maintenance of symptom resolution and healing of erosive esophagitis; in combination with amoxicillin and clarithromycin for treatment of Helicobacter pylori infection and duodenal ulcer disease to eradicate H. pylori ; reduction in occurrence of gastric ulcers associated with continuous NSAID therapy in patients at risk of developing gastric ulcers; long-term treatment of pathological hypersecretory conditions, including Zollinger-Ellison syndrome.

As an alternative to oral therapy when oral therapy with Esoral delayed-release capsules or suspension is not possible or appropriate for the short-term treatment of GERD with erosive esophagitis.

Unlabeled Uses

Non-GERD dyspepsia; Barrett esophagus; stress ulcer prophylaxis.

Esoral is in a group of drugs called proton pump inhibitors. Esoral decreases the amount of acid produced in the stomach.

Esoral is used to treat symptoms of gastroesophageal reflux disease (GERD) and other conditions involving excessive stomach acid such as Zollinger-Ellison syndrome. Esoral is also used to promote healing of erosive esophagitis (damage to your esophagus caused by stomach acid).

Esoral may also be given to prevent gastric ulcer caused by infection with helicobacter pylori (H. pylori), or by the use of nonsteroidal anti-inflammatory drugs (NSAIDs).

Esoral is not for immediate relief of heartburn symptoms.

Esoral may also be used for purposes not listed in this medication guide.

Esoral is a compound that inhibits gastric acid secretion and is indicated in the treatment of gastroesophageal reflux disease (GERD), the healing of erosive esophagitis, and H. pylori eradication to reduce the risk of duodenal ulcer recurrence. Esoral belongs to a new class of antisecretory compounds, the substituted benzimidazoles, that do not exhibit anticholinergic or H2 histamine antagonistic properties, but that suppress gastric acid secretion by specific inhibition of the H ATPase at the secretory surface of the gastric parietal cell. By doing so, it inhibits acid secretion into the gsatric lumen. This effect is dose-related and leads to inhibition of both basal and stimulated acid secretion irrespective of the stimulus.

Indications:

Adult: PO Erosive oesophagitis 40 mg once daily for 4 wk, up to 8 wk if needed.

Maintenance: 20 mg/day. Peptic ulcer 20 mg twice daily for 7 days. NSAID-associated ulceration 20 mg once daily for 4-8 wk. Zollinger-Ellison syndrome Initial: 40 mg twice daily. Usual range: 80-160 mg/day. Gastro-oesophageal reflux disease 20 mg once daily for 4 wk. IV Gastro-oesophageal reflux disease 20 or 40 mg once daily for ≤10 days. Convert to oral therapy as soon as possible.

Esoral is a gastric medicine. It is used to treat conditions caused by excessive acidity in your stomach, such as stomach ulcers and reflux disease (reflux is also known as "heartburn", in which food or acid from your stomach backs up into your mouth, leaving a sour or bitter taste). Esoral treats these conditions by reducing the amount of acid your stomach makes. Esoral may also be used together with other medicines to treat a specific type of gastric problem called Helicobacter pylori or H. pylori gastritis.

Oral Erosive oesophagitis

Adult: 40 mg once daily for 4 wk, extended for another 4 wk if necessary.

Maintenance: 20 mg once daily.

Child: <20 kg: 10 mg once daily for 8 wk; ≥20 kg: 10-20 mg once daily for 8 wk. Hepatic impairment: Severe impairment (Child-Pugh class C): Not more than 20 mg daily. Oral Peptic ulcer

Adult: 20 mg bid for 7 days or 40 mg once daily for 10 days given as a triple therapy with amoxicillin and clarithromycin. Hepatic impairment: Severe impairment (Child-Pugh class C): Not more than 20 mg daily. Oral NSAID-associated ulceration

Adult: 20 mg once daily for 4-8 wk. Hepatic impairment: Severe impairment (Child-Pugh class C): Not more than 20 mg daily. Oral Zollinger-Ellison syndrome

Adult: For short term treatment only. 20 or 40 mg once daily for ≤10 days. Convert to oral therapy as soon as possible. May be administered by inj over at least 3 minutes, intermittent infusion (10-30 minutes) or continuous infusion (over up to 72 hr). Hepatic impairment: Severe impairment (Child-Pugh class C): Not more than 20 mg daily. Reconstitution: Each single-use vial contains 20 or 40 mg of esomeprazole. For IV inj: Reconstitute each vial with 5 ml of normal saline. For IV infusion: Reconstitute each vial with 5 ml of normal saline, lactated Ringer's inj or dextrose 5% inj, then further dilute the resultant solution to a final volume of 50 ml.

Oral Erosive oesophagitis

Adult: 40 mg once daily for 4 wk, extended for another 4 wk if necessary.
