Locion EPC - Pharmacology: Directly dilates vascular smooth muscle by mechanism possibly related to blockade of calcium uptake or stimulation of catecholamine release; reduces elevated systolic and diastolic BP by decreasing peripheral arteriolar resistance; triggers sympathetic, vagal inhibitory, and renal homeostatic mechanisms, including increased renin release, which results in increased cardiac rate and output, and fluid retention; stimulates hair growth by unknown mechanism, but likely is related to its arterial vasodilating action.

Indication: Oral

Management of severe hypertension associated with target organ damage in patients who have failed to respond to max doses of a diuretic plus 2 other antihypertensive drugs.

Locion EPC is a vasodilator that relaxes (widens) blood vessels and improves blood flow.

Locion EPC is used to treat high blood pressure (hypertension). It is usually given together with two other medications including a diuretic (water pill).

Locion EPC may also be used for other purposes not listed in this medication guide.

Locion EPC is a drugs for the treatment of hair loss. This medication has a stimulating effect on hair growth in men and women with androgenetic alopecia (male pattern baldness). The appearance of signs of hair growth observed after 4 months or more of taking the drug 2 times / day. The beginning and the severity of the effect may vary in different patients. After the abolition of Locion EPC new hair growth stops, and after 3-4 months may restore the original appearance.

The exact mechanism of action of Locion EPC as a hair growth stimulant in patients with androgenic alopecia is unknown.

When used systemically this medicine has a vasodilating action. In controlled clinical studies after topical usage Locion EPC in patients with normal as well as with high blood pressure was not observed systemic side effects associated with systemic absorption of this drug.

Locion EPC is an orally effective direct acting peripheral vasodilator that reduces elevated systolic and diastolic blood pressure by decreasing peripheral vascular resistance. Locion EPC is also used topically to treat androgenetic alopecia. Microcirculatory blood flow in animals is enhanced or maintained in all systemic vascular beds. In man, forearm and renal vascular resistance decline; forearm blood flow increases while renal blood flow and glomerular filtration rate are preserved. The predominant site of Locion EPC action is arterial. Venodilation does not occur with minoxidil; thus, postural hypotension is unusual with its administration. The antihypertensive activity of Locion EPC is due to its sulphate metabolite, Locion EPC sulfate.

Oral Severe hypertension unresponsive to standard therapy

Adult: In conjunction with a β-blocker or methyldopa, and a diuretic: Initially, 2.5-5 mg daily; gradually increase at intervals of at least 3 days to 40 or 50 mg daily depending on response. Max: 100 mg daily. Give daily dose in 1-2 divided doses. For a more rapid control of BP: Dose changes may be made every 6 hr with careful monitoring.

Child: In conjunction with a β-blocker or methyldopa, and a diuretic: <12 yr: Initially, 100-200 mcg/kg daily; increase in increments of 100-200 mcg/kg at intervals of at least 3 days, until BP is controlled. Max: 1 mg/kg or 50 mg daily.

Elderly: In conjunction with a β-blocker or methyldopa, and a diuretic: Initially, 2.5 mg daily; increase gradually. Renal impairment: Initiate at smaller doses or at longer dosage intervals. Topical/Cutaneous Male pattern baldness

Adult: Apply 1 ml of a 2% or 5% solution to the scalp bid. 4 mth of treatment may be necessary.

Adult: PO Severe HTN unresponsive to standard therapy W/ a β-blocker or methyldopa, and a diuretic: Initial: 2.5-5 mg/day, up to 40 or 50 mg/day. Max: 100 mg/day. Dose changes may be made 6 hrly (w/ monitoring) for rapid BP control. Topical Male pattern baldness As 2% or 5% soln: Apply 1 mL twice daily.
