Lorazepam Ranbaxy is a benzodiazepine. It works by slowing down the movement of chemicals in the brain. This results in a reduction in nervous tension (anxiety) and causes little sedation.

Pharmacology: Potentiates action of GABA, resulting in increased neuronal inhibition and CNS depression, especially in limbic system and reticular formation.

Indication: Treatment of anxiety, anxiety associated with depression (oral); preanesthetic medication for sedation/anxiety and decreased recall, status epilepticus (IV).

Unlabeled Uses

Relief of chemotherapy-induced nausea and vomiting; acute alcohol withdrawal; psychogenic catatonia.

Lorazepam Ranbaxy is an anxiolytic drug (tranquilizer), a derivative of benzodiazepine. This medication has anxiolytic, sedative, hypnotic, anticonvulsant, muscle relaxant central, antiemetic effect. The mechanism of the anxiolytic, sedative and hypnotic action is associated with increased inhibitory effect of GABA in the CNS. Anticonvulsant action, apparently in part due to increased presynaptic inhibition; inhibited the spread of epileptogenic activity arising in the epileptogenic foci in the cortex, thalamus and limbic structures, but not removed the excited state of the hearth. Muscle relaxant effect is primarily associated with the suppression of spinal polysynaptic afferent pathways and, presumably, an inhibition of monosynaptic afferent pathways; possibly also a direct inhibitory effect on motor nerves and muscle function.

Lorazepam Ranbaxy, a benzodiazepine not transformed to active metabolites, is used to treat anxiety, status epilepticus, and for sedation induction and anterograde amnesia.

Lorazepam Ranbaxy helps to relieve anxiety, especially if it is accompanied by problems sleeping (also known as insomnia). It is also used to help you relax before surgery and dental procedures. Lorazepam Ranbaxy may also be used together with other medicines to reduce nausea and vomiting caused by chemotherapy.

Oral Anxiety

Adult: Usual dose: 1-6 mg daily in 2 or 3 divided doses. Largest dose taken at night. Up to 10 mg daily has been used.

Elderly: Initial dose of 1-2 mg daily in 2 or 3 divided doses. Adjust as necessary. Oral Insomnia associated with anxiety

Adult: 1-4 mg as a single dose given at bedtime.

Elderly: 1-2 mg initially, adjust as needed. Oral Premedication in surgery

Adult: 2-3 mg given the night before the operation followed by a smaller dose if necessary, the next morning. Alternatively, 2-4 mg is given 1-2 hr before the operation.

Child: 1 mth-12 yr: 0.05-0.1 mg/kg (max: 4 mg) given at least 1 hr before surgery. Same dose may be given the night before either as an addition to or to replace the pre-operative dose. Oral Prophylaxis of nausea and vomiting associated with cytotoxic therapy

Adult: For moderately emetogenic chemotherapy: May add 1-2 mg to the antiemetic therapy with domperidone or metoclopramide. Parenteral Acute anxiety

Adult: 25-30 mcg/kg repeated every 6 hr if necessary. Dose may be given via IV or IM inj. Give IV inj at a rate of not >2 mg/minute into a large vein.

Child: Usual: 50 mcg/kg every 4-8 hr. Intravenous Status epilepticus

Adult: 4 mg injected slowly, may repeat once after 10 minutes if seizures recur. Dose should be given at a rate not >2 mg/minute into a large vein.

Child: Neonates and children up to 12 yr: 0.1 mg/kg (max: 4 mg) as a single dose, may repeat once after 10 minutes if needed. Parenteral Premedication in surgery

Adult: 50 mcg/kg, to be given 30-45 minutes before the operation if given via IV inj or 1-1.5 hr before operation if given via IM inj. Intravenous Sedation in critical care

Adult: 0.02-0.06 mg/kg every 2-6 hr as inj or 0.01-0.1 mg/kg/hr as continuous IV infusion.

Child: ≥2 mth: 0.025-0.05 mg/kg (max 2 mg) every 2-4 hr as intermittent IV infusion or 0.025 mg/kg/hr (to a max of 2 mg/hr) as continuous IV infusion.

Oral Anxiety

Adult: Usual dose: 1-6 mg daily in 2 or 3 divided doses. Largest dose taken at night. Up to 10 mg daily has been used.

Elderly: Initial dose of 1-2 mg daily in 2 or 3 divided doses. Adjust as necessary. Oral Insomnia associated with anxiety

Adult: 2-3 mg given the night before the operation followed by a smaller dose if necessary, the next morning. Alternatively, 2-4 mg is given 1-2 hr before the operation.

Child: ≥5 yr: 0.05-0.1 mg/kg to a max of 4 mg given at least 1 hr before surgery. Same dose may be given the night before either as an additive or replacement dose. Oral Prophylaxis of symptoms in antineoplastic-induced emesis

Adult: 2.5 mg the evening before and straight after initiation of chemotherapy. Alternatively, 0.5-2 mg every 4-6 hr. Intravenous status epilepticus

Adult: 4 mg injected slowly (2 mg/min) repeated once if required after 10-15 min.

Child: 0.05-0.1 mg/kg Intravenous Sedation in critical care

Adult: 0.02-0.06 mg/kg every 2-6 hr as inj or 0.01-0.1 mg/kg/hr as continuous IV infusion.

Child: ≥2 mth: 0.025-0.05 mg/kg (max 2 mg) every 2-4 hr as intermittent IV infusion or 0.025 mg/kg/hr (to a max of 2 mg/hr) as continuous IV infusion. Intravenous Prophylaxis of symptoms in antineoplastic-induced emesis

Adult: 1.5 mg/m 2 (max 3 mg) by IV inj over 5 min, admin 45 min before initiation of chemotherapy. Parenteral Acute or severe anxiety

Adult: 25-30 mcg/kg repeated every 6 hr if necessary.

Child: Usual: 50 mcg/kg every 4-8 hr. Parenteral Premedication before surgery

Adult: IV admin: Usual initial dose: 0.044 mg/kg (up to a max of 2 mg) 15-20 min before surgery. A higher dose at 0.05 mg/kg (max: 4 mg) may be used in patients whereby a higher possibility of lack of recall for perioperative events would be beneficial. IM admin: 0.05 mg/kg (up to a max of 4 mg) 2 hr before surgery.

Premed in surgery 2-3 mg the night before operation, then a smaller dose in the morning if needed. IV Status epilepticus 4 mg, repeat once after 10-15 mins if needed. Sedation in critical care 0.02-0.06 mg/kg 2-6 hrly as inj or 0.01-0.1 mg/kg/hr as continuous infusion. IV/IM Acute anxiety 25-30 mcg/kg 6 hrly if needed. Premed in surgery 50 mcg/kg, to be given 30-45 minutes before the operation if given via IV inj or 1-1.5 hr before operation if given via IM inj.
