Pentoxifilina - Pentoxifilina changes the shape of red blood cells in your blood. This makes it easier for these blood cells to fit into small arteries (blood vessels).

Pentoxifilina is used to improve blood flow. Improved blood flow helps to reduce leg cramps and other symptoms of vascular disease (disease related to veins and arteries).

Pentoxifilina may also be used for purposes other than those listed in this medication guide.

Pharmacology: Improves blood flow by decreasing blood viscosity.

Indication: Intermittent claudication on basis of chronic occlusive arterial disease of limbs.

Unlabeled Uses

Treatment of psychopathological symptoms in patients with cerebrovascular insufficiency; treatment of diabetic angiopathies and neuropathies, transient ischemic attacks, leg ulcers, sickle cell thalassemias, strokes, high-altitude sickness, asthenozoospermia, acute and chronic hearing disorders, severe idiopathic recurrent aphthous stomatitis, eye circulation disorders, and Raynaud phenomenon.

Pentoxifilina, a synthetic dimethylxanthine derivative structurally related to theophylline and caffeine, is used in the treatment of peripheral vascular diseases and in the management of cerebrovascular insufficiency, sickle cell disease, and diabetic neuropathy.

Oral Peripheral vascular disease

Adult: As modified-release formulation: 400 mg tid (normally for at least 8 wk), may reduce to 400 mg bid if GI or CNS adverse effects occur. Renal impairment: Moderate impairment (CrCl about 60 ml/min): 400 mg bid; severe impairment (CrCl about 20 ml/min): 400 mg once daily; may reduce further to 400 mg once every other day if needed. Hepatic impairment: Dose reduction may be needed in severe impairment. Special Populations: Adjust dose in patients with impaired renal function or severe hepatic impairment.

Oral peripheral vascular disorders

Adult: As modified-release formulation: 400 mg tid (normally for at least 8 wk), reduce to 400 mg bid if adverse effects occur. CrCl (ml/min) Dosage Recommendation <30 30-50% dose depending on individual tolerance. Hepatic impairment: Severe impairment: Reduce dose.

Indications: Peripheral vascular disease.

This medication is used to improve the symptoms of a certain blood flow problem in the legs/arms (intermittent claudication due to occlusive artery disease). Pentoxifilina can decrease the muscle aching/pain/cramps during exercise, including walking, that occur with intermittent claudication. Pentoxifilina belongs to a class of drugs known as hemorrheologic agents. It works by helping blood flow more easily through narrowed arteries. This increases the amount of oxygen that can be delivered by the blood when the muscles need more (such as during exercise) thereby increasing walking distance and duration.
